Although several efforts have been made to develop therapeutic modalities to inhibit angiogenesis, currently available anti-angiogenic therapies are not always effective, and development of new therapies against angiogenesis is urgent. Previously, we demonstrated that spleen cells in mice stimulated with lipopolysaccharide contain angiogenic cells that can form new blood vessels, and thus, this assay has been developed as a novel circulating angiogenic cell (CAC)-based assay. Human CACs were prepared by the same method as that used in mice, and their proliferative capacity was examined in 3,3'-diaminobenzidine (DAB)-stained rat aortic rings cultured in the presence of conditioned medium from human breast cancer cell lines. Conditioned media from breast cancer cell lines stimulated the proliferation of CACs derived from human peripheral blood. This assay is more sensitive and has a higher specificity than the conventional tube-based CAC assay. However, when human CACs were cultured with conditioned medium from human breast cancer cell lines in the presence of the drugs, there was no effect on cell proliferation, even if the tumor growth-promoting factors in conditioned medium were blocked. This is the first report of an experimental system to evaluate the effect of drugs on angiogenic factors, and these results suggest that the CAC-based assay is suitable for studies of tumor angiogenesis, including anti-angiogenic therapy.Q:
